Iontophoresis is used for non ionized drugs required in a small dosage. Trans- dermal administration can be passive or facilitated. In passive administration, the non-ionized drug traverses the skin through the stratum corneum. The skin, being a semi permeable membrane, allows only a small amount of any drug molecule to passively penetrate the skin. Ionized drugs do not easily penetrate this barrier and are not suitable for routine trans-dermal delivery unless an external source of energy is provided to drive the drug across the skin. Facilitated diffusion can use electrical, iontophoresis energy. In iontophoresis, this external source of energy is in the form of an applied direct electrical current. Iontophoresis is a non-invasive method of delivering drugs into the body. With a low-level electrical current, iontophoresis repels drug ions through the skin and into underlying tissue. The use of iontophoresis for transdermal drug delivery has been successfully used by medical professionals for decades and continues to increase dramatically because of its advantages
Sonophoresis is a Low-frequency ultrasound can increase the transdermal delivery of many drugs, including macromolecular drugs. The main mechanism is ultrasonic cavitation. Most researchers pointed out that it could change the form-structure of stratum corneum keratinocytes and, in this way, it can improve the permeability of skin. Low-frequency sonophoresis has been in use for in-vitro experiments and in-vivo animal experiments, and so far, both small-molecules transdermal delivery and macromolecules transdermal delivery have been successfully performed in many experiments
